Does the first attack against a target get mastery bonus or do you have to ‘prime it’?

That's why you start out with FSK which counts as the first hit boiiiiiiiiiiiiiiiiiiiiiii

Just start with FSK and then pop Serenity.

As long as it wasn't a repeat of the last ability, yes. Leaving or entering combat doesn't reset the tracking of Mastery.
