I think you mean transmute master.
anyways, click on the cancel button (red circle with a line) next to your elixir master description. That should remove the specialization, and then you should be able to learn your new specialization (If I recall correctly
)

You can unlearn your specialization by clicking the 'cancel' sign on your professions.
There is no "Gem Master" spec so... cant help you there.

Keep in mind that learning a new specialization is now rather expensive, four truegold bars are needed to learn transmutation for example.
