Yes. The heart is an actual item on his corpse that you need to loot.

I'm pretty sure this is a yes, but you would have to go back and do Lei Shen a second time...say if your last runestone came off of Lei Shen.

If you park yourself at the black prince before starting the run, then if you get a runestone at an earlier boss you back port out hand in q/get followup then port back before Lei Shen.
