The maximum distance for DisplayPort at full bandwidth is 3 meters. At higher distance you'll lose bandwidth, which means you can't go as high resolution or refresh rate anymore.
So the answer to your question: it depends what resolution/refresh rate you expect to get through there.

There are 50 foot display port cables so... maybe? at 1440p 60hz... maybe not? Hard to say for sure I'd look around tho I'm sure someone has had to do it before.
Yes you can. You can buy cables with boosters in them but they are expensive.

Is it going to a different room or something? There's no way that the computer itself could exist near the screen, and the mouse/keyboard/etc could use longer cables instead?
If the tower can live near the screen, then you can just get a USB hub and powered USB extension lead and use that to connect peripheral. Probably a fair bit cheaper than DisplayPort over the same distance.
